鹿港天后宮湄洲媽祖香火鼎盛，更因廟宇年代久遠而規模宏偉，聞名遐邇，尤其建築結構富麗堂皇，古色古香，雕樑畫棟獨具匠心，彩繪及木石雕刻，皆精緻絕倫，巧奪天工，故有藝術殿堂之稱。廟中陳列的珍貴史料及宗教文物，更是令中外人士嘆為觀止。如前清皇帝的御筆匾額，文武百官的獻匾，古代碑記及本宮昔日往湄洲祖廟謁祖照片和祖廟贈與的大靈符，聖母寶璽，均是本省絕無僅有之文物。此外，尚有明朝宣德年製的進香正爐，清朝雕製的精緻鳳輦，全副儀杖等，數量之多，難以枚舉，自從湄洲祖廟毀於文革後，奉祀於本宮之湄洲開基聖母寶像，及有關湄洲文物，更受到廣大信徒重視，而成為碩果僅存的歷史瑰寶。
